1.Nowadays pen drives has become a common way to carry info .This has become a boon as well as bane .Whenever we insert a  pendrive autoplay window pops up .if some malicious code is attached to AUTORUN.INF which is responsible for autoplaying as soon as we click on ok our system gets infected.
To disable autplay in windows xp  follow these steps  either hold shift while the device is inserted or goto run typein gpedit.msc
which stands for gpedit(group policy editor).msc(microsoftconsole) .Goto administrative templates >System Tools.There u will see a option called "Turn off autoplay" enable it to turn off autoplay or using regedit goto HKEY_LOCAL_MACHINE\System\Currentcontrolset\services\USBSTOR.
Double click on the entry named start in the right pane.The default value in the Value data field will be 3 change this value to 4 to disable the pendrive.

To display autoplay in vista follow these steps
a)open your control panel,now click on "Play CDs or other media automatically" to open the autoplay dialogue .NOW uncheck the box to disable autoplay on all kinds of devices.
b)Invoke gpedit.msc thru run now browse to windows components\Autoplay policies
change the value of "turn off autoplay" to enable

2) Never open EMAIL attachments that end in .vbs or .js, since Virus are commonly spread with these files typical user would never have a reason to open these files.

3)never accept or run files from untrusted or unknown users.

2)using sandbox

Deep freeze is an excellent tool to protect ur system from virus .This works different from antivirus .When u install this small tool it asks us to choose the drives that we want to freeze.After selection is made it records the state of the system Which is coined as freezing .whenver we restart the system the drive/system is restored to its original state.Let us take an example a friend of us inserted an infected drive into our system.dont worry just restart ur system,Its that easy.but if u wanna update ur system or install new softwares then you have to disable it get the updates now install it.so may get a doubt that virus may enter while disabling it . i Would recommend to download a trial version of antivirus scan the drive and enable the freeze option .This is far better than installing the antivirus completely.This software is best suited to internet cafe where after installation u will not update frequently.
